Sir Alexander Fleming – the discoverer of Penicillin
Great Britain History is made up of very renowned scientists from all their history. This is me decide to write about one of the most famous British scientist – Sir Alexander Fleming discovered penicillin. The discovery of penicillin was more than a purely random event.
Alexander Fleming's discovery of penicillin is one of the most celebrated event of an accident in the science. In the conventional story, was a stray mold spore carried by an open window and landed on an exposed bacterial culture, after Fleming a clear zone where the bacteria was slain struck, he immediately recognized the therapeutic significance of the event, and it was only a matter of time before penicillin was a miracle. Fleming often emphasized the role of chance in his work. Despite the many awards and awards he received, he was fond of reminding others, "I did not invent penicillin. Nature did. I only discovered by accident."
There was even more "likely" to the story often told, however. Moreover, the traditional account obscures a considerable amount of scientific work that the efficacy of penicillin identified as an antibacterial agent. Without a number of researchers, who aggressively pursued the potential in the first observation Fleming, penicillin would probably not be become a "discovery" on this occasion. The fuller story suggests a more complex view of science – as supported by both the risks of the circumstances and targeted effort by the researchers.
Renewed interest in the history of Fleming's work began many years ago when a noted bacteriologist in London the windows of Fleming's lab at St. Mary's Hospital were so constructed that they could not open. How could a stray mold spore have wandered in, not even by chance? Second, he noted, Penicillium spores will not germinate under the conditions described by Fleming. Someone else then noted that the particular species of Penicillium would probably not be suspended in the air of London. Although common bread mold is a variety of Penicillium, was much rarer P. penicillin notatum Fleming's produced.
The most likely source of the mold, it now appears, was down from a mycology lab Fleming. There were probably on track throughout. Furthermore, Fleming never known for cleanliness in his lab. Open cultures would not be uncommon. It seems almost inevitable, that the mold would infect a culture of its past or later.
The conditions of infection would be important. Fleming believed, based on his earlier work on lysozyme, that penicillin acted by lysing bacteria to enter. This would certainly account for the watery appearance of the area in its culture where the bacteria were absent. In this case the spore, just need to land on the culture plate – and this is how Fleming's own casual observation reported. But we have learned that penicillin works by blocking the synthesis of chemicals used by bacteria to build cell walls. Penicillin not necessarily kill bacteria. Rather, it prevents their effective reproduction. A spore landing on an existing culture would probably be no immediate discernible effect. The fungus would have had to establish themselves first when it comes to preventing the further growth of bacteria. Temperature, while Fleming was away on vacation from his lab have allowed this, or inoculated Fleming may have a plate that was already moldy. In both cases, a stray mold spores alone would not have created what Fleming observed.
The fact that Fleming noticed that the original culture also seems unlikely. Fleming was not in the holes as the template effect routine-ly study of its cultures, although to hear him when he returned from his one month summer vacation in 1928. In fact, he had waived the now famous culture and allowed to soak in a bucket of lysol. A former member of his lab, however, stopped by to visit, and Fleming made him different cultures. Among these he selected passing the critical culture from the top of the waste pile where he had escaped liquid disinfectant. Only then was Fleming struck by the unusual pattern of growth. He was clearly impressed, but because he showed the culture of many colleagues throughout the day and went to some of the foreign anti-bacterial properties saw he studies.
Fleming was not the first scientist to have noticed that the antibacterial activity of fungi. In 1871 was Joseph Lister (famous for introducing the practice of antiseptic surgery) showed that a template in a sample of urine appeared to inhibit bacterial growth. In 1875 John Tyndall reported to the Royal Society in London, that some sort of Penicillium bacteria had caused some of his cracks. In 1877 Louis Pasteur and Jules Joubert noted that the air micro-organisms growth of anthrax bacilli in the urine, which was previously sterilized braking.
Most dramatically, Ernest Duchesne had completed a doctoral dissertation in 1897 on the evolutionary competition between microorganisms, with emphasis on the interaction between E. coli and Penicillium glaucum. Duchesne reported how the mold, the bacteria in the culture eliminated. He also inoculated animals with both the form and a lethal dose of typhoid bacilli, indicating that the fungus so that the animals from contracting typhoid. He urged more research, but went into the army following his degree and died of tuberculosis before ever returning to the investigation. Chance, here, worked against his discovery (Potential or discovery?) To pay off.
Several other researchers – almost certainly unknown Fleming – had noticed the effects of Penicillium fungi to bacteria. Fleming was not unique in this respect. But notice a phenomena not necessarily mean that it will be followed. The Fleming's chance in case maybe less the appearance of the moldy culture itself that Fleming had a habit of pursuing strange phenomena. Fleming pursued his observation.
Yet Fleming not follow through on his "discovery" in ways that we can expect, knowing that the current role and importance of penicillin. Fleming originally observed the action of penicillin in 1928. Yet he failed to start clinical trials. Nor has he strongly supports the use of penicillin in the treatment of humans until 1940. The events during break these twelve years are perhaps the most telling in the history of penicillin.
Fleming was definitely looking for antibacterial agents in 1928 and he studied the potential of penicillin. But he was not impressed. He found that penicillin was not toxic to animals and that it is not harmful white blood cells (leukocytes), but he found that penicillin would not be recognized if taken orally. Penicillin taken by injection, alternatively, was excreted in urine in a matter of hours – long before could the consequences. For Fleming, penicillin was the therapeutic options are limited, perhaps to topical antisepsis.
Fleming did continue to use and advocate penicillin in the first years after its discovery. But he saw the value of penicillin in the first place in the context of bacteriology. Penicillin suppresses the growth of certain bacterial species, that a selective culture specific others (such as that caused influenza, acne and whooping cough). In this role penicillin became a valuable tool in the manufacture of vaccines – a Fleming important task administered St. Mary's Hospital. Production of penicillin continued on a weekly basis throughout the year 1930, but all for the purification of bacterial cultures. The penicillin was rough – good enough for Fleming's goal, but hardly strong enough for a serious infection in humans to destroy. Meanwhile, Fleming turned his study to another group of chemical bactericides, the sulfonamides.
The exercise of penicillin in the treatment of human infections was due to end another lab, led by Howard Florey in Oxford. In 1938 Ernst Chain, Florey's an employee, began a search for natural anti-bacterial agents, as part an attempt to under-stand their mechanisms more completely. He chose to study three, among them penicillin. Fleming 1929 paper offered a thread of the information chain can achieve up, but with a very different purpose. By early 1939 Florey and Chain began to suspect the medical potential of penicillin. But they could not just testing: penicillin was difficult to produce and purify. Florey had trouble finding funding. By that time, the war in Britain meant that additional funds were not available for exploring some possibilities. Aid finally arrived in late 1939 by the Rockefeller Foundation in the U.S.
Florey shift resources in his department to penicillin project. Before they demonstrate the efficacy of penicillin, they had several technical challenges. They were validated to improve, to refine a test for determining the strength their extracts, and the volume of production. After five months of work – in May, 1940 – they had enough of the brown powder to test for mice. Penicillin allowed several mice injected with lethal doses of virulent streptococci to survive. The potential of penicillin to treat infections proven or seemed real. Florey and Chain repeated their tests as a double-check, and then went in the right dosages and duration of treatment to adopt, publish their results in August.
But the study hardly done. The results would transfer to humans? To know, they had to scale production yet again. Based on relative weight, a person would need about 3,000 times the penicillin used by a mouse. And commercial support was not forthcoming. Labs in Oxford, biscuit tins and bottles used for the mold cultures gave way to hundreds of bedpan-like ships are stored on a bookshelf. Purification was found from the laboratory to dairy equipment. Column chromatography allowed the group to the relevant groups and isolating the concentrate their solutions. All this was in the service of a clinical trial. – And after the first test in early 1941, they had returned to their methods as a way to remove impurities that found had caused side effects. The tests went quite well, but it had asked two professors, five and ten graduate assistants working almost every day of the week for some months to produce enough penicillin to treat six patients.
Fleming took note of the striking results. But he did not disturb his research agenda. He knew the value of penicillin was still in the research of economic mass production. Thus, the research – and in a sense, the discovery – was not completed. Florey took his case to America again, where work began on the scale breweries. One of the main technical assistant was a new medium for the mold cultures, increase yields tenfold. Other pharmaceutical companies in Britain were now interested, but the volume of production was somewhat limited at first. After half set of clinical trials in 1942-43, but production began in earnest. In half years, the industry would produce enough to treat 200 persons per month. Two years later the United States produce enough to quarter one million to treat patients per month.
Many scientists, Fleming among them, were convinced that the provision of the chemical structure of penicillin would enable chemists to produce it synthetically and more economical. Once the structure was determined, however, synthesis appeared at least as expensive as extraction. The "failure" seems an exception This story is different graced by luck. But not all research ventures pay off as expected – a chance works in both directions in science.
Nobel Prize winner Peter Medawar once commented, "I was disappointed that the traditional story of the discovery of Fleming not stand up to a critical eye, because I would have liked to have believed it's true, but even if it was true, it would not have told us much about the efficacy of happiness. "Here, Medawar referred to the substantial work that one lucky event turns into a real discovery. There is more to knowing that what meets the eye. First, we must acknowledge and be prepared to assess the significance of a result of the comments turn. Fleming had a habit of playing in the lab and playing with idiosyncrasies. He pursued a chance phenomenon that even his colleagues unimportant, even to guess without ultimate meaning.
Furthermore, the importation of an observation is not always clear. Chain and Florey recognized a therapeutic potential in which Fleming saw it but vague. And they were willing to invest resources to continue. Fleming, Florey and Chain all shared in the Nobel Prize for Medicine in 1945. Their joint award reminds us that the discovery of penicillin was more than a purely random event.
